Testing Principle
Viruses are non-cellular microorganisms composed mainly of nucleic acids and proteins. They can only replicate within living cells, are extremely small in size (10–30 nm in diameter), and can pass through bacterial filters. In cells infected by certain viruses, round or irregular bodies of varying sizes and quantities, known as viral inclusion bodies, can be observed under a light microscope. RNA viruses typically form cytoplasmic inclusion bodies, while DNA viruses form nuclear inclusion bodies. Some viral inclusion bodies that are difficult to observe usually require special staining methods for confirmation. The modified Macchiavello method uses basic fuchsin and methylene blue for staining, which provides good coloration for RNA coronavirus inclusion bodies. Basic fuchsin readily binds to eosinophilic inclusion bodies in the cytoplasm, staining them red. Differentiation with an acidic solution effectively removes non-specific staining from other tissue components. Methylene blue counterstains the background blue.


Stain Results

RNA viral inclusion bodies: Appear red within the cytoplasm of lung tissue cells.

Red blood cells: Appear pink or purplish-blue.

Fibrin exudates: Appear light pink.

Cell nuclei and connective tissue: Appear blue.
